Původ Pátého pluku se datuje k Dělnicko-rolnickým antifašistickým milicím (MAOC), které vytvořila Komunistická strana Španělska a které sloužily jako ochrana socialistických a komunistických vůdců, složených převážně z ozbrojenců z JSU. Když v roce 1936 vypukl státní převrat, byly tyto rozděleny do pěti praporů milicí, které se aktivně účastnily obrany Madridu. Pátá z těchto kolon byla konstituována autonomně jako 5. pluk lidové domobrany. Pátý pluk podílel 20. července 1936 na obsazení kláštera salesiánů v Madridu, a přemenil ho na výcvikovou základnu na vojenský výcvik dělníků a rolníků, a se proslavil díky náročností ve vojenském výcviku a soudržnost jejích členů pod velením, které nedávalo možnost zpochybňovat jejich rozkazy. Počet narukovaných mužů rychle rostl s tím, jak se rozvíjely vojenské akce v okolí hlavního města, v období od srpna do listopadu se zvýšilo z 6 000 na 20 000 milicionářů. Do konce července již minimálně 1000 příslušníků odešlo na frontu, přičemž velká část zůstala v týlu pod výcvikem nebo na jiných misích. Pluk byl téměř vždy v čele, v hlavních akcí začátku španělské občanské války , aby se zabránilo pádu republikánského hlavního města Madridu , účast v bitvách Somosierra , Guadarrama , Talavera a Toledo, navíc k evakuaci muzea z Prado do Valencie. V republikánské armádě byly některé smíšené brigády, jako 1., 10., 31. nebo 68., které byly z velké části tvořeny jednotkami 5. pluku. Dá se říci, že 5. pluk se stal jedním ze vzorů pro formování Republikánské lidové armády a mnozí z jeho milicionářů přešli k vytvoření prvních smíšených brigád. Vyšli z ní význační vůdci Republikánské lidové armády jako Juan Guilloto León „Modesto“, Enrique Líster , Valentín González „El Campesino“ a Etelvino Vega Martínez . Například 11. republikánská divize (nějaký čas pod velením Enrique Lístera ) vznikla začleněním 5. pluku do Republikánské lidové armády.. Definitivně přestal fungovat 22. ledna 1937. Vittorio Vidali , jeden z jeho velitelů, toho dne prohlásil: "Pátý pluk je mrtev! Ať žije lidová armáda!" Nebyl pouze vojenskou jednotkou, protože jeho činnost se rozšířila i do dalších oblastí, rozvíjela důležitou sociální a kulturní práci, podporovala rozvoj vzdělanosti a kultury mezi nižšími vrstvami v boji proti negramotnosti. Známé byly jeho plakáty a nástěnné malby, jeho putovní knihovny a „divadelní partyzáni“, které vytáhl do ulic a na frontu. U pátého pluku byli zařazeni nebo spřízněni s komunistickými básníky a spisovateli známými jako Rafael Alberti, Cesar Arconada, Pedro Garfias, Miguel Hernandez, José Herrera Peter , María Teresa León popř.Juan Rejano. Stejně tak inženýři a architekti, jako Luis Lacasa Navarro nebo Manuel Sánchez Arcas, lékaři jako Juan Planelles Ripoll , 10 profesorů jako Josep Renau , Wenceslao Roces nebo Alberto Sánchez a malíři jako Alberto Sánchez Pérez .
