At the time of the Japanese attack on Pearl Habor, the U.S. Air Force possessed a fairly extensive collection of types that could only be considered obsolete. In a frantic effort to speed up war readiness, the Air Force began to pull everything off the production lines that had wings and could be at least marginally used for military service. Thus, in early 1942, 24 Douglas DC-2s entered service and were taken over by the Air Force from civilian carriers. These risers were designated C-32A because, while they were not the result of any development or order based on the prototype XC-32, the Army needed to get them into its marking system. Most underwent only minor modifications and retained their original civilian engines, but some received the Army's Wright R-1820-33.

The other five of these stands weren't even pure DC-2s because shortly before, in late 1941, their original operator TWA had traded these machines to Northeast Airlines for three new DC-3s. The new user swapped the wings with those of the DC-3, hence the nickname DC-Twee for these five standees. They were subsequently taken over by the Army and, notwithstanding their new wings, were attached to the other DC-2s under the same designation.

The C-32A was used in a variety of roles, for cargo transport, staff and communications purposes, and training. Some of them didn't even really make it into military hands, their transfer was a paper affair. Similarly, in contrast, the Army itself handed over some C-32As to private carriers to strengthen the transport network, which as a whole also served the Army's interests. Five stands were even taken over from the RAF and served in Europe, two stands from the Australian Air Force and flew in the Pacific.

All C-32As lived to see the end of the war, when they were sold off as surplus to private hands or ended up in scrap.
